Microbial Fermentation: Development & Scale-Up
This course emphasizes extensive laboratory work and demonstrations in microbial fermentation from bench scale to pilot scale. Laboratory experiments are supported by lectures and discussions on the fundamentals of microbial growth, growth kinetics, strain development, and scale-up strategies. Participants will gain the skills and knowledge to design and optimize fermentation processes in batch and fed-batch cultures.
Day 1
Lecture:- Overview of microbial fermentation
- Kinetics of fermentation
- Media formulations for fermentation processes
- Recombinant protein production in bench-top fermentors
- Fermentor setup and sterilization
- Fed-batch strategy and experimental design
- Inoculum development, fermentor inoculation, pH and DO preparation, and calibration
Day 2
Lecture:- Protein expression in bacteria
- Computer control of fermentation
- Design of bench top fermentors
- Continuation of bench scale fermentation-harvest
- Oxygen mass transfer in 10-liter fermentor
Day 3
Lecture:- Scale-up strategies in fermentation processes
- Biomass monitor
- Anaerobic fermentation
- Pilot scale (100L) fermentors
- Fermentor setup, sterilization and preparation for inoculation
- Fermentation monitoring and control
- Troubleshooting issues
Day 4
Lecture:- Strain development and improvement
- Harvest and processing of 100-liter fermentors
- Continuous flow centrifugation
